Dosage ; Adults: Chemotherapy and radiotherapy -
highly emetogenic regimens; 8mg by slow 1.V inj immediately
prior to treatment followed by dose of 8mg by 1.V inj
repeated 2-4 hours for 2 doses
(or by 1mg per hour by 1V. inf. for up to 24hrs) then 8mg orally BD
for up to 5 days.
Post-operative-prophylaxis 8mg orally 1 hour prior to
anaethesia followed by two doses or 8mg orally at 8hrly intervals
or single dose of 4mg by slow 1.v inj at induction of anaesthesia.
Treatment single dose of 4mg by slow 1.V inj.
Children: 5mg/m2 1.v infusion immediately prior to chemotherapy
followed by 4mg orally BD for up to 5 days.

| Mode Of Action | A specific serotonin (5HT3) antagonist (Emetogenics like cytotoxic drugs and radiation damage gastrointestinal mucosa. This leads to the release of serotonin from the enterochromaffin cells of the gastrointestinal tract. 5-HT3 receptors are stimulated causing transmission of sensory signals to the vomiting center via vagal afferent fibers to induce vomiting. Granisetron binds to the 5-HT3 receptors hence blocking vomiting mediated by serotonin release). |
