Promethazine

Dosage ; Allergic or anaphylactic reactions, oculogyrosis,

crisis of Parkinsons syndrome: up to 50mg deep I.M. I.V

(with other agent: 25-50mg slowly).

Pre-medication in anesthesia: Adult- 25-50mg 1-2 hours before surgery,

children 12-16yrs - 12.5-25mg, younger children less.

Night sedation and insomnia: 25mg at bedtime.

Increased to 50mg when required, 5-10 years; 20-25mg at bedtime,

2-5years; 15-20mg, under 2 years not recommended.

Nausea, vomiting, vertigo, labyrinthine disorders, motion sickness:

For HCl salt Elixir: 25mg at bedtime on the night before traveling.

Repeat before traveling when required,

5-10yrs; 10mg at night and the following morning,

2-5yrs; 5mg at night and the following morning.

For theocrate that acts longer than HCl salt.

Dose: 25-75mg (max. 100mg) daily.

Motion sickness: 25mg at bedtime on night before traveling or 25mg 1-2hrs

before traveling. Vomiting in pregnancy (severe):

25mg at bed- time (max 100mg daily)

Mode Of Action

Antihistamine (H1)
Drug Indication Allergic or anaphylactic reactions, oculogyrosis, crisis of Parkinsons syndrome, Pre-medication in anesthesia, Night sedation and insomnia, Nausea, vomiting, vertigo, labyrinthine disorders, motion sickness and Vomiting in pregnancy (severe)
Precautions Prostrate hypertrophy; narrow angle glaucoma; and urinary retention.
Contra-Indications Premature infants; acute asthmatic attack; and epileptic; SC injection
Side Effects Drowsiness; psychomotor impairment; anti-muscarinic effects (e.g. urinary retention; dry mouth; GI disturbances; blurred vision); allergic reactions; epileptic form seizure in patients with cerebral cortex focal lesion; muscle weakness; tachycardia; tight chest; paradoxical CNS stimulation - mostly in children; thrombophlebitis for IV use.

Special Information It is safe in early pregnancy but it is postulated to have a potential of causing neurological disorders in infants.
