| Mode Of Action | Artemisinin derivatives are concentrated in the parasitized erythrocytes and it is believed that iron from ingested haemoglobin of the parasite?s victim reduces ?endoperoxide bridge? releasing a highly reactive free radical iron (IV) oxo species that rips apart the parasitic cells. |

| Precautions | Use with caution in patients with severe hepatic and renal insufficency, patients refusing food, during second and third trimester of pregnancy and lactation. Adequate contraception is required. Due to risk of dizziness, fatigue / asthenia, driving and use of machinery is not recommended. Not indicated for prophylactic use. Not recommended for severe malaria. Not to be used concurrently with other anti-malarials unless no other treatment option. Special care with patients previously treated with halofantrine. |

| Side Effects | Headache, dizziness, abdominal pain, anorexia, sleep disorder, palpitation, cough, diarrhoea, vomiting, nausea, pruritus, rash, arthralgia, myalgia, asthenia, fatigue, somnolence, involuntary muscle contractions, paraesthesia, hypoesthesia, abnormal gait, ataxia, hypersensitivity, personality disorders, asymptomatic QTc prolongation (no causal relationship confirmed). |
| Dosage | (Based on 20mg/120mg tablets) Adults: 4 tablets stat, then 4 after 8hrs, then 4BD x 2/7 (24 tablets). 5-15kg: 1 tablet stat, then 1 after 8 hrs, then 1 BD x 2/7 (total = 6 tablets). 15- 25kg: 2 tablet stat, then 2 after 8hrs, then 2 BD x 2/7 (total = 12 tablets). 5- 15kg: 3 tablet stat, then 3 after 8hrs, then 3 BD x 2/7 (total = 18 tablets). In the event of vomiting within I hr of administration a repeat dose should be taken. |
