| Mode Of Action | Griseofulvin inhibits fungal mitosis by disrupting the mitotic spindle through interaction with polymerized microtubules |
| Drug Indication | Fungal infections of skin, hair and nails. |
| Precautions | Severe skin reactions (e.g. Stevens-Johnson syndrome, toxic epidermal necrolysis) and erythema multiforme; discontinue if severe skin reaction occurs. Elevations in AST, ALT, bilirubin, and jaundice; discontinue if jaundice occurs. |
| Contra-Indications | Pregnancy, avoid during pregnancy or within 1month of treatment, men should not father children within 6 months of treatment. |
| Side Effects | GI disturbances, skin rashes, photosensitivity, dizziness, fatigue, oral contraceptives failure, blood dyscrasia, breakthrough bleeding, confusion and impaired judgment. Hepatotoxicity, exacerbation of SLE, oral thrush. |
| Dosage | 500mg - 1000mg daily. Tinea capitis: 4-6wks. Tinea corporis 2-4weeks. Onychomycosis of finger nails: at least 4 months. Onychomycosis of toe nails: at least 6 months. Children: 10mg/kg/day. |
