Mode Of Action

ivermectin binds selectively to glutamate-gated chloride ion channels in invertebrate muscle and nerve cells of the microfilaria. This binding causes an increase in the permeability of the cell membrane to chloride ions and results in hyperpolarization of the cell, leading to paralysis and death of the parasite. It also acts as an agonist of the neurotransmitter gamma-aminobutyric acid (GABA), thereby disrupting GABA-mediated central nervous system (CNS) neurosynaptic transmission.

Drug Indication Onchocerciasis, Strongyloidiasis, Bancroftian filariasis and Scabies
Contra-Indications Hypersensitive to any components of the formulation.
Side Effects Ophthalmologic effect like limbitis, punctate opacity, conjunctivitis, and eyelid edema; Mazzotti-like reaction; pruritus; tachycardia; facial edema; headache; peripheral edema; diarrhoea; dizziness; skin rash; itching; postural hypotension; anorexia; somnolence; tremor.
Dosage Onchocerciasis: 150mcg/kg as a single dose. The dose may be repeated every 3 to 12 months, depending on the recurrence of symptoms and/or microfilariae. Strongyloidiasis: 200mcg/kg as a single dose. Bancroftian filariasis: 20 - 200 mcg/kg as a single dose. Scabies: 150 mcg/kg as a single dose for one treatment (repeated after 2 weeks in immunocompromised patients).
